Rapid anticancer drug screening using patient-derived lung cancer organoids: a case report

چکیده
A variety of anticancer drugs and targeted agents for lung cancer have been developed, but some patients do not respond to these medications as intended. Therefore, there is an urgent need develop a tool predicting the drug response each patient. Patient-derived tumor organoids (PDOs) emerged reliable in vitro tools developing precision medicine. Herein, we describe case 50-year-old nonsmoking man who was diagnosed with non-small cell cancer. Initially, no clinical symptoms were found this Postoperative pathology confirmed stage Ib tumor, epidermal growth factor receptor (EGFR) mutation (exon21p.L858R) detected patient’s specimen. The patient later showed intracranial relapse 17 months after complete resection. An organoid culture established from resected brain metastatic tissue sensitivity test showed, within 72 hours, that resistant gefitinib osimertinib. Our results recapitulated drugs, demonstrating potential PDOs

Targeting mutant RAS in patient-derived colorectal cancer organoids by combinatorial drug screening
Colorectal cancer (CRC) organoids can be derived from almost all CRC patients and therefore capture the genetic diversity of this disease. We assembled a panel of CRC organoids carrying either wild-type or mutant RAS, as well as normal organoids and tumor organoids with a CRISPR-introduced oncogenic KRAS mutation. Using this panel, we evaluated RAS pathway inhibitors and drug combinations that ...
متن کاملRapid Revival of a Patient after very Severe Metabolic Acidosis: A Case Report
Background: Metabolic acidosis is a fatal finding in trauma patients that complicates the process of resuscitation. Case: The case was a 37-year-old man with open fracture in both legs and fracture in second lumbar vertebral (L2). The serial arterial blood gas (ABG) test results showed a pH value of 6.7 indicating a very severe and special case of metabolic acidosis. The rate of mortality f...
متن کاملA Case Report of Small Cell Lung Carcinoma in a Patient with Progressive Diffuse Sclerosis
Connective tissue diseases, including scleroderma, may be associated with an increased risk of lung cancer. The most common type of lung cancer associated with systemic sclerosis is adenocarcinoma or bronchoalveolar carcinoma, and small cell carcinoma is rare. This reports presents a 54-year-old non-smoker woman with a history of scleroderma who was examined due to aggravation of shortness of ...
متن کاملHepatic organoids for microfluidic drug screening.
We introduce the microfluidic organoids for drug screening (MODS) platform, a digital microfluidic system that is capable of generating arrays of individually addressable, free-floating, three-dimensional hydrogel-based microtissues (or 'organoids'). Here, we focused on liver organoids, driven by the need for early-stage screening methods for hepatotoxicity that enable a "fail early, fail cheap...
